Why it matters · written by this site

A reference work rather than a result: it reports the affinity of twenty-five psychedelics at fifty-one receptors, transporters and ion channels assayed by the NIMH Psychoactive Drug Screening Program, and adds literature data on ten more p. Its argument is that the selectivity usually assumed for these compounds is not there — the drugs surveyed interact with forty-two of the forty-nine sites broadly assayed, and the paper proposes that this diversity of receptor interaction is what underlies the qualitative diversity of the drugs themselves p. It also introduces a normalisation that factors potency out of a Ki so that two compounds' multi-receptor profiles can be compared as shapes rather than as magnitudes p. It is the second-largest single contributor of measurements to this archive a.

Verified Abstract read in full via PubMed (PMID 20126400, DOI 10.1371/journal.pone.0009019); the paper is open access in PMC2814854 but the full text was not read. Every claim above is stated in the abstract, including the counts twenty-five, fifty-one, ten, forty-two and forty-nine. The normalisation method is described in the abstract as introduced there; how it works was not examined. Nothing is claimed about the method's reception or use elsewhere.

Not supported Whether the normalisation has been adopted by others, and whether the selectivity claim is now the consensus. Neither is in the abstract.

Receptor binding profiles and behavioral pharmacology of ring-substituted N,N-diallyltryptamine analogs

Klein LM, Cozzi NV, Daley PF et al. · Neuropharmacology 2018 · PMID 29499272

    40 measurements at 24 targets · Measurements

    5-MeO-DALT, DALT

Why it matters · written by this site

A study that puts binding and behavior side by side rather than inferring one from the other. Ten ring-substituted N,N-diallyltryptamines were assayed at forty-five receptor and transporter sites and separately tested in mice for the head twitch response, a 5-HT2A-mediated behavior used as a proxy for hallucinogen effect p. The finding that matters is a negative one: head twitch potency did not correlate with either 5-HT2A or 5-HT1A binding affinity on its own, and only a regression treating the two together accounted for it, with 5-HT2A contributing positively and 5-HT1A negatively p. The authors read this as 5-HT1A activation buffering the effect of tryptamine hallucinogens p. It is the largest single contributor of measurements to the archive's DALT records a.

Verified Abstract read in full via PubMed (PMID 29499272, DOI 10.1016/j.neuropharm.2018.02.028). The forty-five sites, the HTR rank order, the absence of a single-receptor correlation, and the signs of the two regression contributions are stated there. The abstract's receptor subscripts are lost in PubMed's plain text -- it renders '5-HT' where the paper prints a subscripted subtype -- so the two subtypes named above are taken from the sentences where the abstract spells them out, and no subtype is asserted from a passage where the digit is missing.

Not supported The R value of the regression, which appears in the abstract as 'R = 0.8729' with the exponent lost, so it cannot be told whether the paper reported R or R². Neither is quoted.

Receptor interaction profiles of novel N-2-methoxybenzyl (NBOMe) derivatives of 2,5-dimethoxy-substituted phenethylamines (2C drugs)

    Rickli A, Luethi D, Reinisch J et al. · Neuropharmacology 2015 · PMID 26318099

    26 measurements at 10 targets · Measurements

    2C-H, 2C-I, LSD, Mescaline

Why it matters · written by this site

The paper that characterised the NBOMe compounds when they were new and their pharmacology was, in its own words, poorly defined p. It measures each 2C phenethylamine beside its N-2-methoxybenzyl analogue and beside LSD at monoamine receptors and transporters, so the effect of the substitution can be read directly: adding the N-2-methoxybenzyl group raises affinity at 5-HT2A, 5-HT2C, α1, D1–D3 and H1 and at the monoamine transporters, while lowering it at 5-HT1A and TAAR1 p. The result is a set of very potent 5-HT2A agonists, EC50 0.04–0.5 μM, resembling LSD in their 5-HT2A and α1 affinity but not in their dopamine receptor binding p. This archive holds both halves of that comparison, which is why the paper appears on several 2C and NBOMe dossiers at once a.

Verified Abstract read in full via PubMed (PMID 26318099, DOI 10.1016/j.neuropharm.2015.08.034). The receptor lists, the direction of each affinity change, and the EC50 range 0.04–0.5 μM are all stated there. The abstract notes that TAAR1 was assayed with rat/mouse receptors rather than human, which is why no claim is made here about human TAAR1.

Not supported The individual Ki values, which are in the paper's tables and not in the abstract. Where this archive shows an NBOMe affinity it comes from the ingested rows, not from this note.

Structural basis of opioid receptor activation by PCP and ketamine

    16 measurements at 4 targets · Measurements

    3-HO-PCP, Ketamine, Phencyclidine (PCP)

Why it matters · written by this site

The reason ketamine's opioid receptor rows in this archive stopped being blank. Before it, every ChEMBL record for ketamine at KOR, MOR and DOR was a screening result quarantined as a non-result — 'Inhibition < 50% @ 10 uM and thus dose-response curve not measured' — so the archive held the question and none of the answer a. This paper measures it: cryo-EM structures of human KOR and MOR bound to (S)-ketamine and to PCP, with binding affinities and cAMP potencies for PCP and all three ketamine forms at all three opioid receptors p. It also reports the ligand-free state of KOR, which serves as a negative control for the ligand densities in the bound structures p. Sixteen of its measurements are held here, transcribed from the paper rather than from a database a.

Verified Full text read from the publisher PDF: Results, Discussion, and the Methods sections covering membrane preparation, saturation and competition binding, the cAMP inhibition assay and the warm water tail withdrawal assay. The quarantine comment quoted above is this archive's own stored `activity_comment` on those ChEMBL rows, not the paper's words. The structures, the apo KOR control and its stated purpose are in the Results. See tools/literature_measurements.json for what was and was not taken from it.

Not supported The paper's stereoselectivity result — that (S)-ketamine binds and activates opioid receptors more strongly than (R)-ketamine — cannot be shown on this site, because the archive holds racemic ketamine and neither enantiomer. That absence is recorded in tools/literature_measurements.json rather than papered over.

Why it matters · written by this site

A brain-imaging study rather than a pharmacological one p. Sixty-two adults with no previous psychedelic experience were imaged with fMRI and 64-channel EEG in four contexts — eyes-closed rest, guided meditation and music, and an eyes-open movie — at a baseline session and again after a single 19 mg oral dose of psilocybin p. What it reports is a redistribution rather than a loss of order: global functional connectivity fell in sensory cortex and rose in associative cortex during the eyes-closed conditions, the gap between eyes-closed and eyes-open connectivity narrowed in every network, and the later EEG block found the same convergence in alpha power p. Low-dimensional embeddings of each participant's dynamics separated the four contexts under psilocybin and not at baseline, and that separation was graded by the participants' own ratings of self-dissolving and boundary-dissolving experience p. Three of the six reported milestones on this archive's psilocin simulation are taken from it — at the two times it states for its scans, and at its day-after follow-up — and it supplies no measurement row anywhere on this site a.

Verified Full text read from the published PDF: article, Methods, the figure and Extended Data captions, and the Reporting Summary. The Supplementary Information and the Extended Data figures themselves were not read, so nothing that appears only there is claimed above. The design is open-label and unblinded at every stage including analysis, with each participant's baseline session about a week earlier as the only comparison — there is no placebo arm, and that is stated here because it is the weakest design of any work this archive draws a time from. Analysed sample sizes vary by condition after quality control (fMRI 56-60 per condition, EEG 58, and 54 for the embedding analyses), so the sixty-two above is the number imaged in both sessions and not the n behind any single result. What the milestones do and do not take from it is recorded in tools/effect_milestones.json.

Not supported Anything connecting these findings to the rest of this site. The paper reports no binding, no occupancy and no plasma concentration, so none of it can be checked against the affinity tables the simulation computes occupancy from, and the milestones drawn from it sit beside the modelled curve rather than being derived from it. Its P values for the network comparisons are computed across cortical vertices rather than across participants and are not quoted anywhere here. Its brain maps are not shown at all: the archive's regional figures are transcript levels from the Human Protein Atlas, a different quantity in a different parcellation, and drawing a connectivity change on them would assert a correspondence nobody has published.

The notes

Who wrote what on this page

The distinction the rest of the site is built on, applied to its own bibliography.

Everything above a landmark note is a record, not a description. The title, journal, year and authors are whatever PubMed returned for the identifier. The counts beneath are this build’s own rows, computed when the page is rendered. Neither is written by hand, and neither says anything about a paper’s quality.

The 5 landmark notes are the exception, and they are the only prose on this site that describes a paper. They carry the same fence as the history notes on the compound dossiers: every factual claim carries a marker — p for the paper as it was read, a for this archive’s own rows — and each note records exactly what was read. For most of them that is the abstract and not the full text, which is stated rather than glossed over, because a note may not claim more than its reading supports.

A note is never written from general knowledge, and there is no note where the reading does not support one. One paper was considered and deliberately left without one:

Molecular biology of 5-HT receptors (Boess & Martin, Neuropharmacology 1994)

It contributes more measurement rows to this archive than any other paper, which is precisely why it is tempting to write a note about it from general knowledge. PubMed returns no abstract for it and nobody here has read the review. A note would therefore be a description of a paper this project has not opened.

Would need The review read, or at minimum an abstract to quote from.

Provenance

How this list is built

Four places in this archive cite a PubMed identifier: assay rows carrying a reference_pmid, the pharmacology passages quoted from PubChem, the toxicology passages, and the curated acute-effects table. All four are gathered here. tools/fetch_literature.py resolves each identifier against PubMed once and stores the bibliographic record — last resolved 2026-09-27.

Quarantined compounds are excluded inside the query rather than after it, so a record the site does not show cannot credit a paper with measurements. That is the same rule every other count on this site follows, and the integrity report says which records it applies to.
